Of course, you have to know C++.
Then ROOT can be seen as a set of Classes for data
analysis/storage/display. Browsing through : 
http://root.cern.ch/root/Reference.html
Will give you an idea of theyr organisation, and provide you an everyday
help for every class interface.

But maybe you should start working with the tutorials :
http://root.cern.ch/root/Tutorials.html
That are also on your disk under $ROOTSYS/tutorials.

Browsing the 'HOWTO's' :
http://root.cern.ch/root/Howto.html
Will certainly answer some of questions you may have when first trying
to really use root to performù some task.

Last but not least, the new (but still in developpment) ROOT user guide:
http://root.cern.ch/root/RootDoc.html
may worth a printout.
